This excellent perspective from AstraZeneca highlights where oncology drug discovery is heading. Thank you, Mark Cobbold and Puja Sapra for articulating a future in which success is driven not by a single modality—whether anti-PD-1, ADCs, T-cell engagers, CAR-T, antibodies, or small molecules—but by rational, precision and combinations tailored to tumor biology. One theme that stands out is the central role of immuno-oncology in these future regimens. While targeted therapies, ADCs, and radiopharmaceuticals can generate meaningful responses, durable tumor control will likely require effective engagement of the immune system. An equally important challenge is the immunosuppressive tumor microenvironment, which continues to limit the full potential of many therapeutic modalities.
