PIK3CA encodes the p110α subunit of PI3K — the most commonly mutated kinase in human cancer. When mutated (E545K, H1047R), it becomes a runaway signal telling cells to grow, divide, and evade death. The good news? Researchers at UT Southwestern and around the world are developing next-generation inhibitors that target only the mutated form — sparing healthy cells and reducing side effects. New clinical trials with RLY-2608 and STX-478 are showing early promise. And data from the INAVO120 trial (NEJM 2025) shows that combining PI3K inhibition with hormone therapy and CDK4/6 inhibitors can meaningfully extend survival. Science takes time.
