More compounds don’t always mean more understanding. In lead optimization, exploring multiple changes at once can make it harder to see what’s actually driving results. Potency shifts, ADME signals move, but the “why” stays unclear. The programs that move forward with more confidence tend to do the opposite. They narrow the question, control the variables, and design each cycle to test one thing at a time.
