Want to complement MALDI imaging of FFPE samples with LC-MS proteomics, but lack the right sample preparation workflow? In this talk, Andreas Sonnen of UMC Utrecht presents how his team tackled this challenge while studying pancreatic neuroendocrine tumors. Starting with MALDI imaging, they identified peptide patterns distinguishing tumor subgroups. Follow-up LCMS analysis of microdissected regions revealed unexpected molecular signatures. This prompted the next question: 𝘊𝘢𝘯 𝘵𝘩𝘪𝘴 𝘩𝘦𝘵𝘦𝘳𝘰𝘨𝘦𝘯𝘦𝘪𝘵𝘺 𝘣𝘦 𝘦𝘹𝘱𝘭𝘰𝘳𝘦𝘥 𝘢𝘤𝘳𝘰𝘴𝘴 𝘭𝘢𝘳𝘨𝘦𝘳 𝘍𝘍𝘗𝘌 𝘤𝘰𝘩𝘰𝘳𝘵𝘴? To do so, they needed a simple and reliable workflow for LCMS proteomics. Using the 𝗕𝗲𝗮𝘁𝗕𝗼𝘅𝗶𝗦𝗧 𝗙𝗙𝗣𝗘 𝘄𝗼𝗿𝗸𝗳𝗹𝗼𝘄 for the first time, they were able to: ✅ Prepare FFPE samples 𝘄𝗶𝘁𝗵𝗼𝘂𝘁 𝗽𝗿𝗶𝗼𝗿 𝗟𝗖𝗠𝗦 𝗲𝘅𝗽𝗲𝗿𝘁𝗶𝘀𝗲 and send them for analysis ✅ Reproduce 𝗯𝗶𝗼𝗹𝗼𝗴𝗶𝗰𝗮𝗹 𝗴𝗿𝗼𝘂𝗽𝗶𝗻𝗴 𝗮𝗰𝗿𝗼𝘀𝘀 𝘁𝗶𝘀𝘀𝘂𝗲 𝘁𝘆𝗽𝗲𝘀 ✅ Generate ~𝟮,𝟬𝟬𝟬 𝘁𝗼 𝟱,𝟬𝟬𝟬 𝗽𝗿𝗼𝘁𝗲𝗶𝗻 𝗜𝗗𝘀 from small sections (0.28–30 mm²) Their results show how accessible workflows can help connect spatial readouts with LC-MS based proteomics, enabling new insights from existing FFPE archives Thank you, Andreas, for this very interesting sneak peak into the world of pathology! ➡️https://ow.ly/Ygir50Z9eRW #FFPE #tissueprep #proteomics #lasermicrodissection #LCMS #sampleprep #spatial #MALDIMSI
