BCL6 TCIPs are highly effective in high-risk models of DLBCL with strong BCL6 expression, including post-CAR T relapse models. But what happens when tumors have mixed patterns of BCL6 expression? Spoiler alert: π·πππππππππ. DLBCLs represent heterogeneous populations of cells with plasticity to move between phenotypes. Under pressure from BCL6-targeting TCIPs, they polarize to an insensitive BCL6-negative phenotype. But this comes along with other targetable dependencies!
