Osivelotor is an oral investigational drug for the treatment of sickle cell disease (SCD) designed to inhibit hemoglobin polymerization. In a phase 1, open-label ADME study conducted in healthy participants at ICON’s clinical research unit in the Netherlands, a single 200 mg dose (including a 14C mictotracer) demonstrated slow elimination, with a half-life of approximately 27–30 days and almost 90% recovery in urine and feces over nearly seven months months with six completers. The treatment was well tolerated, with the drug predominantly circulating unchanged in whole blood and glucuronide metabolite presence in plasma. These findings provide important insights into osivelotor’s absorption, metabolism, and excretion, supporting its continued development as a potential treatment for SCD.
