Today is International Migraine Awareness Day! Migraine is a prevalent neurological disorder affecting over one billion people worldwide. It is characterised by recurrent attacks of moderate to severe headache, often accompanied by nausea, sensitivity to light (photophobia) and sound (phonophobia), and occasionally visual or sensory disturbances known as aura. The pathophysiology of migraine is complex and not yet fully understood. It involves dysregulation of brain networks involved in pain and sensory processing. Key mechanisms include cortical hyperexcitability, activation of the trigeminovascular system, and the release of inflammatory neuropeptides such as calcitonin gene-related peptide, or short CGRP. Emerging evidence suggests that the endocannabinoid system may also play a role in migraine pathophysiology. Proposed mechanisms include a deficiency in endocannabinoids such as anandamide, which may contribute to increased CGRP levels, and the modulation of cortical excitability through inhibition of glutamatergic transmission. For a detailed review, see Lochte et al. (2017). https://lnkd.in/ewZvT-Zf Medical cannabis shows promising therapeutic value for the treatment of Migraine: Observational and cross-sectional studies have reported improvements in migraine frequency, sleep, and reduced use of conventional analgesics with medical cannabis (e.g., Aviram et al., 2020; https://lnkd.in/e9mCmzSe). Although the current evidence supporting the use of phytocannabinoids in migraine is promising, it remains preliminary and large-scale randomized controlled trials are still needed to confirm the efficacy of cannabinoid treatment in Migraine. #MigraineAwarenessDay #Migraine #MedicalCannabis #Cannabinoids #CGRP #EndocannabinoidSystem #Neurology #CannabisResearch #PainManagement #Phytocannabinoids #ClinicalResearch #Cannaflos
