Developed by NIST to support standardization of extracellular vesicle reference materials, this study used four orthogonal techniques, Cryo-EM, particle tracking analysis, asymmetrical flow field-flow fractionation, and microfluidic resistive pulse sensing to evaluate particle size distributions and concentrations of MSC and LNCaP prostate cancer cell EVs. Quantitative discrepancies in particle number concentrations reached up to two orders of magnitude across methods, while RNA-seq revealed notable inter-laboratory variation, emphasizing the need for reproducible and harmonized EV characterization methods.
