For large-scale manufacturing of longer peptides, should we pursue linear SPPS or conjugation of SPPS-based fragments? This is a frequently asked question in peptide manufacturing. Yet peptide length alone should not determine the synthesis strategy. In our recent case study, we evaluated linear and fragment SPPS for a commercially relevant GLP-1 receptor agonist and found that fragment synthesis is not automatically more efficient or cost-effective for longer peptides. Instead, the optimal approach depends on molecule-specific factors, process performance, and overall efficiency.
