Excited to share our development of a novel and remarkably simple approach to SMLM called "FILM" that can reach down to molecular resolution. FILM is based on gradual, flow-based delivery of conventional fluorescent probes (e.g. antibodies used for IF, antibody fragments, fluorescent phalloidin) to a sample under gentle LED Illumination on a standard widefield microscope. Gentle Illumination allows for the recording of roughly 100x more photons from each fluorophore-labeled probe than standard SMLM approaches, allowing for nm and even sub-nm resolution in situ at room temperature. See the link below for our bioRxiv preprint, which has not yet been peer reviewed. https://lnkd.in/e_uEb8zs
