Multiplexing with primary antibodies from the same species has always been a challenge. The ๐ฃ๐ฟ๐ฒ๐บ๐ถ๐ &๐ฆ๐๐ฎ๐ถ๐ป protocol with Smart Secondary Nanobodies addresses this directly. And here is a 5-color example showing what that looks like in practice. Three targets were labeled using mouse primary antibodies, detected with Smart Secondariesยฎ (๐๐น๐๐ผ๐ง๐ฎ๐ดยฎ-๐ซ๐ฎ ๐ฎ๐ป๐๐ถ-๐บ๐ผ๐๐๐ฒ ๐๐ด๐๐ญ) plus direct GFP tag labeling for a fourth channel via ๐๐น๐๐ผ๐ง๐ฎ๐ดยฎ-๐ซ๐ฎ ๐ฎ๐ป๐๐ถ-๐๐๐ฃ. ๐ฃ๐ต๐ฎ๐น๐น๐ผ๐ถ๐ฑ๐ถ๐ป ๐ฆ๐ง๐๐ฅ ๐๐๐จ๐ serves as a structural counterstain for actin. โฌActin: abberior STAR BLUE Phalloidin ๐ฅNUP target 1 (GFP-tagged): FluoTagยฎ-X2 anti-GFP abberior STAR 550 ๐ฉGolgi: FluoTagยฎ-X2 anti-mouse IgG1 abberior STAR ORANGE ๐ฆNUP target 2: FluoTagยฎ-X2 anti-mouse IgG1 abberior STAR RED ๐งMitochondria: FluoTagยฎ-X2 anti-mouse IgG1 abberior STAR deepRED These images showcase two new additions to the nanobody conjugate portfolio, developed in collaboration with NanoTag Biotechnologies: ๐ฆ๐ง๐๐ฅ ๐ฑ๐ฒ๐ฒ๐ฝ๐ฅ๐๐ and ๐ฆ๐ง๐๐ฅ ๐ฑ๐ฑ๐ฌ conjugates extend the spectral range of the FluoTag toolbox. #SmartSecondaries #PremixAndStain #STARdyes #FluoTag #abberior #NanoTagBiotechnologies #MulticolorImaging
