Cell Engineering & Protein Expression •Engineered Sf9 insect cells as a platform for recombinant protein expression by achieving efficient CRISPR-mediated gene integration. •Identified and deleted genomic coding sequences of the in Sf9 cells to accelerate cell engineering. •Tested operational parameters, feeding strategies, and media to optimize protein expression in Sf9 cell lines using perfusion processes and maintained it using aseptic techniques for 36 days. •Purified protein using affinity liquid chromatography and membrane filtration. •Expressed HLA-DR cell surface receptor (key role in immune system) using baculovirus expression vector system in Hi5 insect cells. •Cloned and maintained DNA sequences in MachI E. coli cell line. •Drafted, edited, and maintained experimental protocols, batch records, and laboratory notebook. •Analyzed and presented data to department and cross-functional teams. High Dimensional Mass Cytometry Analysis •Developed comprehensive graphical user interface to facilitate analysis of high dimensional mass cytometry data using statistical models and machine learning. •Analysis of large mass cytometry datasets using a pipeline of quality control, image processing (MCDViewer), cell segmentation (CellProfiler), single cell analysis, and spatial analysis (histoCAT). •Automated redundant or error-prone tasks by developing custom MATLAB codes. •Collaborated with the Michigan Medicine Departments of Dermatology resulting in 5 co-authored papers on several diseases of the skin (HS, PPP, PP, cSLE, JDM, AD). •Validated a panel of 36 cancer and immune marker antibodies to use in mass cytometry analysis of metaplastic breast cancer.
