Worked in Dr Kaustuv Datta lab : I screened for temperature sensitive mutants of yor205c,which is a GTPase by nature and has its role in ribosome biogenesis in yeast mitochondria particularly with assembly of small subunit. I found some putative ts mutants which stops growing on plate containing glycerol as a sole carbon source when shifted to non permissive temperature. The mutation occurred in the strains is very crucial for identifying domains of yor205c which is responsible for its binding to smaller subunit of ribosome or keeping ribosome intact as a whole. Yeast utilizes mitochondrial TCA cycle to obtain energy when grown on glycerol so cells with non functional mitochondria in case of ts mutants will stop to show growth on glycerol containing media.
