Specifically, I have a mastered skill-set in the following areas: - Protein Expression/Fermentation across multiple cell lines (E.Coli, HEK293, CHO, and hybridoma) at many scales ranging from 1mL to 60L - Setup and Handling of TFF equipment both automated and manual operation. - Protein Capture and Cleanup with IMAC, HIC, AEX/CEX, Protein A/G/L, and other industry standard methods Utilizing AKTA instruments and unicorn 5/7. - Protein characterization across upstream and downstream process via SDS-PAGE, CE-SDS, DLS and Protein Solubility. - Operation of Agilent HPLC equipment/software in both reverse phase and standard SEC mode for aggregate measurements and stability - Operation of BLI (octet) equipment/software for both protein kinetics and titer measurement with extensive development time on both areas. - Waters LC-MS QTOF with UNIFI systems for reduced mass glycan analysis. - Technical Operations: Microsoft products, computer equipment, lab equipment. If something stops working I am generally the person asked to fix it. My career goal is to lead a team where I can leverage my leadership skills as well as my technical expertise to drive product innovation at the support or discovery stage. Patients need therapies, and passion at all levels makes this happen. Outside of my work life I have a very important job: I am a father and a husband. I also have many hobbies involving computer building/tinkering, cooking, exercising and the occasional book on astrophysics by Sagan or Tyson. While I am very driven in my career, balance between life and work is incredibly important to me.
Manufacturing Science and Technology Downstream Group Leader
Leading the Pilot Lab downstream team to deliver GLP Tox. and Rep. Batches of biologics at the 50L - 1000L scale with tight deadlines and challenging modalities. Leading multidisciplinary technical teams and driving successful campaigns from transfection to IND. Deliver GLP/GMP process development improvements, mentor junior level scientist on implementation strategies. Interrogate and resolve GLP/GMP process characterization and impurity challenges and present findings across the organization. Provide on floor support, person-in-plant, batch record drafting/review, and tech transfer.
Leading GLP Tox and Pre-IND programs through the Scale-Up Lab to deliver representative batches of biologics at pilot scale. Acting as downstream lead in multidisciplinary technical team meetings. Interfacing with process development to drive downstream tech transfer, process fit, batch record drafting and review.
Assisting in the biochemical development and production of an oxygen carrying protein therapy into clinical trials. Key Accomplishments: -Developed a number of small scale assays/purification schemes to test and verify protein expression. -Developed an assay which monitored protein solubility across upstream production. -Worked as the primary analytics during fermentation process development with a CMO. -Developed the upstream scale up of 12L to 60L in house production a day. -Developed various protocols and protocol improvements to current processes. Worked directly with PI to study the effect of specific site mutations on the AMPA receptor, to learn more about the biophysical process of its operation. Key Accomplishments: -Created 19 mutant constructs to be used for study by the lab. -Optimized various protocols Responsibilities: -PCR Mutagenesis -Cell Culture -Cloning -mRNA work, specifically transcription and microinjection -Animal surgery and post surgery care Administrative Responsibilities -Work with regulatory departments to demonstrate that current protocols and regulations are being followed. -Handle appropriate training of any new lab members in any of my general responsibilities. -Maintain accurate records of all transactions and procedures.
