Development & Validation of an LC-MS/MS Method for Iohexol Measurement University of Manchester Masters Project: Developed and validated a liquid chromatography–tandem mass spectrometry (LC-MS/MS) method for the quantification of iohexol in human plasma to enable gold-standard measurement of glomerular filtration rate (GFR). This work supports more accurate kidney function assessment in patients where creatinine-based eGFR is unreliable. Aims included: • Optimised sample preparation using protein precipitation with 0.1 M zinc sulphate in 70% methanol/water for improved recovery and matrix effect reduction. • Established calibration and validation parameters in line with bioanalytical method validation guidelines, assessing linearity, limit of detection, limit of quantitation, accuracy, precision, and stability. • Operated advanced analytical instrumentation including Waters Acquity–Xevo TQ-S LC-MS/MS with ZSpray ionisation and Acquity I-Class UPLC. • Performed data processing using MassLynx v4.1 to generate calibration curves, evaluate QC sample performance, and assess analytical robustness. • Investigated potential interferences and assessed carryover, recovery, and reproducibility to ensure clinical suitability of the method.
