    Development of a fermentation process of plasmids at 10L scale : The production of plasmids is a limiting step in the processes using transient transfection in mammalian cells for recombinant production and viral vectors but also to produce DNA vaccines and plamids for gene therapy. The eventual aim of this project is to develop a process to produce R & D batches of purified plasmids at a 10l scale. The availability of this method internally would remove this limitation, at least for development stages. Activities : - Search and study of the litterature in the field of plasmid production with E.Coli. - Establishment of Batch and fed-batch process to develop high yield production process with Sartorius Biostat - BPlus bioreactor at 2L scale/Optimisation of processes - Establishment of analytics tools (Molecular Biology) In parallel, cell culture studies at small scale (T-Flasks, Shake-Flasks ans bioreactor).     R&D studies with the low field NMR technique on food and cometic products : Low-field NMR characterizes the compartmentalization of protons and the texture of samples. The obtained data after direct analysis or confrontation with other biological parameters will identify quality markers and improve the manufacturing process or the product formulation. During this internship, I was in charge of several projects of low-field NMR on products in food and cosmetic industries. The first selected study for this report deal with the preservation of an emulsion and the second determine the influence of raw material nature on chocolate syrups texture.
