My research centered around Mycobacterium tuberculosis and the role of transcription factors in the resistance of antibiotics. It was supervised by Konstantin Brodolin in the Bacterial Enzymes and Antibiotics Resistance team at the Institut of Research on Infectiology of Montpellier. - Resolved two structures of Mycobacterium tuberculosis RNA polymerase by CryoEM at less than 2.4Å nominal resolution - Purification of 10 recombinant proteins involved in the transcription by FLPC - Numerous biochemical analyses such as transcription assays involving handling radioactive samples, fluorescence polarization, microfluidic diffusional sizing or more regular assays (EMSA, KMnO4, DNAse footprinting...) - Screening the activity of antibiotic derivatives. Work published: Jung et al. 2024, Angewandte Chemie - Writing of a scientific manuscript article as a first author (in progress for publication). - Shared my work as oral presentation and posters at international congress
- Part of the fertility screen project among Plasmodium berghei - Engaged in rodents handling and various procedures (anaesthesia, malaria parasites injections). - Installation of the Celsee and first trials of single-cell analysis
- Complete setup and management of a new insectary with Anopheles stephensi rearing. - Communication coordinator within the new laboratory team and Sanger Institute. - Transfert of the PlasmoGEM ressource counting more than 5 000 clones.
- Training in Anopheles stephensi breeding and writing of insectary management protocols. - Engaged in cell culture and Plasmodium mutagenesis through PlasmoGEM ressources.
Supervised by O. Grépinet and I. Virlogeux-Payant in the Lab Team SPVB (Signalisation Portage et Virulence Bactérienne). Subject : Regulation study of Rck by the nucleoprotein H-NS among Salmonella Typhimurium. - Mutagenesis by Datsenko and Wanner’s method and transductions. - Western-blots anti-3xFLAG and anti-6xHis with Stain-Free® or classical SDS-PAGE gels. - Development of a semi-quantification normalised on total proteins via Photoshop. - Purification of the nucleoprotein H-NS by affinity chromatography. - Team work, critical analysis improvement and thesis essay.
