Characterisation of hydrophobic variants and specific protein modifications
pharmaceutical development for mAB´s (Biosimilars) current postion as drug product project team member (DP-STM) responsible for a mAB project: preparation of development report and conduct lab scale studies. the main focus is on process characerization related topics (e.g. scale down models, formulation robustness studies) lab and commercial scale related troubleshooting (e.g. define and ensure a robust manufacturing process) support and perform technology tranfer to commercial site: preparation of process flow charts, manufacturing instructions, process validation related documents
Protein production: recombinant protein production by fermentation (10L bioreactors, mineral salt media), Protein purification: soluble and in-soluble & native and denatured state utilizing IMAC, HIC, SEC, Refolding of proteins purified under denaturated conditions Perform detergent screenings in order to increase solubility and stability of proteins, DSC, fluorescence and circular dichroism, artificial chaperone system used for renaturation Artifical membrane system: liposomes and nanodiscs used to stabilize GPCRs class B receptos establishing reconstitution protocols for different type of membran proteins (GPCRs, Transporter, Kinases) Bioactivity: ligand binding studies with various methods e.g. radio ligand, ITC, pulldown assay, Biacore
