Performed qualification of scientific equipment like cryogenic liquid nitrogen freezers, refrigerators, incubators and gas analyzers. Designed and executed protocols for facilities validation. Designed and implemented ADME/PK program for Immunology/transplantation and CNS projects for small molecules at the discovery stage to evaluate potential for drug development. Represented DMPK functions on drug discovery project teams. Designed and conducted more than 50 DMPK profiles of small molecules for transplantation, auto-immune diseases and CNS including ADME in vitro and PK studies. Developed and present recommendations based on data evaluation and interpretation to internal teams and senior management in US and Japan.
Provided DMPK support for all Transplantation and CNS projects (at this location): - ADME profile: microsomes stability, CYP inhibition (DDI), Solubility, Absorption, Protein Binding - Analysis and interpretation of ADME data for choosing pre-lead compounds - PK prior all testing in vivo to find an efficacious dose (according to in vitro data) in efficacy models. - PK/PD analysis Lead the bio-analytical lab for drug discovery: -Developed and validated LC/MS/MS methods for drug metabolism and PK profiles Directed a team (2 people) responsible for the development and implementation of new technologies to enhance drug metabolism studues supporting discovery projects; special emphasis on intestinal absorption, hepatic clearance and drug transporters. Measured induction of CYP enzymes by RT-PCR in human and rat primary hepatocytes. Wrote IRB protocol to obtain specimen for ADME research projects
Analyzed metabolites of synthetic oxysterols (compounds to inhibit cholesterol biosynthesis) in vitro and in vivo. Isolated intestinal microsomes and performed in vitro assays with inhibitors of ACAT an enzyme thought to regulate cholesterol levels. Adapted analytical methods for complete separation and identification of sterol precursors in Smith-Lemli-Opitz syndrome.
