Absorption Distribution Metabolism Excretion Department •Developed models and methods, standardized and analyzed In vitro microsomal metabolic stability of more than 600 new chemical entities (NCE’s) using standard compounds with different animal species like human, mouse, rat, monkey, dog, mini-pig which was helpful for the organization to identify the nature of target NCE in humans. •Developed methods for determination of Phase-II conjugation reactions like glucuronidation, sulfation, acetylation, glutathione with S9 fractions of different animal species which again supported discovery of NCE. •Standardized and executed enzyme inhibition assays of CYP450 enzymes involved in drug metabolism using Spectrofluorimeter and HPLC. Determination of Ki using Dixon, Eadie hoftees plot. •Performed and coordinated with Bioanalytical department for studies like Metabolite profiling and CYP450 reaction Phenotyping which provided the important information of which particular CYP450 enzyme mainly responsible for drug metabolism. •Conducted validation of Cell Culture based - Human Hepatocyte CYP450 Induction assays which involved isolation and purification of mRNA, preparation of cDNA and thereby study of gene expression by Real time PCR. •Developed In vitro permeability models (hexadecane and lecithin PAMPA). •Literature survey for setting up model for CaCO2 permeability. •Performed studies on NCE to identify its penetration of Blood Brain Barrier with use of Evans blue marker. •Coordinated and interacted with different departments at all stages of drug discovery for the selection of NCE. •Involved in data interpretation and analysis of In vitro-In vivo correlation of ADME parameters. •Monitored that all the ADME parameters meet the requirements and proper selection of NCE. •Supervised, managed and trained lab assistant, college student and junior colleagues.
