I'm a protein biochemist who is versatile in all ascpects of protein preparation, characterization and crystallization. I can rely on a broad methodical knowledge in protein analytics, molecular biology, cell culturing, or FPLC-based purification. I'm familar with prokaryotic and eukaroytic expression systems as well as a wide range of biophysical methods for protein characterization and was able to solve various protein structures in projects with different backgrounds.
Besides other proteins relevant for infection processes, my work focuses on the structural characterization of biosynthesis pathways of large versatile natural compounds with therapeutical potential. For this purpose, the proteins are recombinantly expressed and purified on modern FPLC systems. Before crystallization, the obtained samples are comprehensively characterized. The obtained structures are interpreted to shed light on the individual synthesis steps.
During my PhD studies, I was working on different human proteins with therapeutical potential and gained first insights into structure-based drug design. The proteins were expressed in eukaryotic as well as prokaryotic expression systems and purified on FPLC systems. Were necessary, refolding protocols were established. Furthermore, the proteins were characterized by various methods including e.g. SDS-PAGE, Western-Blotting, DLS or thermophoresis. Subsequently, crystallization trials were performed to obtain structures of the apo as well as of the ligand-bound states and the respective structures were determined and intensively interpreted.
