the project was to unveil the function of a microRNA containing gene silencing complex in regulating developmental processes in plants. Biochemical-, genetics- and molecular biology methods were performed. Based on the observation that a protein of a silencing complex (AGO7) is present in two different compartments in the cell, I found that AGO7 migrates differently in 2D gels, when extracted from different compartments. Consequently, I performed Co-IP assays with antibodies against tagged AGO7 protein from both compartments. The Co-IPs were analysed by mass spectrometry. I was using bioinformatic tools (MaxQuant/Perseus) to quantify and statistically analyse the outcome of repetitive Co-IP experiments. I could identify several potential candidates that interact with AGO7 and function in gene silencing. These candidates are currently further studied. The project was part of the Exzellenzcluster CellNetworks at the University of Heidelberg.
