I lead a group of 18 highly qualified personnel (Scientists, Associate and Assistant Scientists and Students) which focus on protein purification, analytics, biophysics and mass spectrometry. We built up from scratch a laboratory where we employ state of the art methods to study protein conformation and dynamics and to analyze protein therapeutics. Our technologies include: Size Exclusion Chromatography (SEC) and Asymmetric Flow Field-Flow Fractionation (AF4) Separation coupled with UV-VIS, Fluorescence, Differential Refractive Index (dRI), Multi Angle (MALS) and Dynamic Light Scattering (DLS) for accurate determination of protein molar mass and size under native conditions and for protein-protein interactions. Circular Dichroism and Fourier Transform Infra-Red Spectroscopy for assessment of protein secondary structure; Differential Scanning Calorimetry to study thermodynamics and protein stability; SDS-PAGE, western blotting and capillary Isoelectric Focusing to assess protein purity, charge heterogeneity and identity; Mass Spectrometry for intact mass measurements, peptide mapping, determination of post-translational modifications and protein-protein interactions. We have extensive experience in pre-development work, manufacturability studies for protein drug candidates and transfer of know-how to development. We focus on a multitude of research projects where we apply Hydrogen Deuterium Exchange Mass Spectrometry (HDX-MS) to study epitope and paratope mapping and protein conformation and dynamics. Our group is characterized by high flexibility and dynamics and is capable to tackle a multitude of tasks from different expertise areas.
