Prof. Dr. Jean Pieters laboratory Focus in biochemistry / immunology / cellular signaling Project: Characterization of candidates identified in the interactome of coronin 1 and investigation of their potential role in T cell homeostasis and survival Main activities: - Cell culturing (Jukat T cells, mouse and human primary T cells) in a BSL-2 environment - Genetic engineering/perturbation using CRISPR/Cas9, generation of knockout cell clones - Plasmid construction, PCR, Gel Electrophoresis, Sanger Sequencing - Flow cytometry, single cell cloning by FACS - Microscopy: Cell imaging using immunofluorescence, co-localization studies of proteins, deconvolution - various biochemical assays (Western Blotting, immunoprecipitation, intracellular antibody-mediated staining, affinity purification, T7E1, peptide competition assay, BCA) - Data analysis with various software's: GraphPad Prism, Geneious Prime, SnapGene, FlowJo, Inkscape, R studio, FIJI, OMERO, Huygens Professional, Microsoft Excel
